Occurrence of Gluconatecobacter Diazotrophicus in the Botanical Seed of Sugarcane

Abstract

Gluconacetobacter diazotrophicus is a natural endophyte of several crops, among them sugarcane, from which it was discovered in Brazil in the decade of 1980, isolated from roots, stems and leaves. Because of its beneficial characteristics: nitrogen fixing, phytohormones producer, phosphorus solubilization and potential biological control, has been deeply investigated. Regarding its systemic character, this work considered its possible natural occurrence in the botanical seed of sugarcane. To accomplish this, total nucleic acids were extracted from seeds obtained from 14 crosses supplied by the National Hybridization Center of Sancti Spiritus. DNA from the total nucleic acids extracted was submitted to 23S RNAr gene PCR amplification by means of specific primers. The expected amplified DNA bands were not obtained indicating the absence of this bacterium in the samples analyzed.
